DESCRIPTION:“Stress for success”: Unlocking Venetoclax sensitivity in T-ALL through the mTORC1–ISR–BMF axis.\n\n\n\nby Prof. Vincenzo Ciminale\, MD\, Istituto Oncologico Veneto\, Padova\, Italy\n\n\n\n\n\nWhen: Thursday\, 13.11.2025\, 14.30–15.30 \n\n\n\nWhere: Aud. 4\, BBB \n\n\n\nHost: Bjørn Tore Gjertsen \n\n\n\nAbstract\n\n\n\nRefractory/relapsed T-cell acute lymphoblastic leukemia (R/R T-ALL) remains a major therapeutic challenge at present. Our recent studies showed that inhibition of mTORC1 markedly sensitizes T-ALL cell lines and patient-derived xenografts (PDXs) to the BCL-2 inhibitor Venetoclax. RNAseq\, gene set enrichment analysis\, and analysis of the temporal trajectories of response to the mTORC1 inhibitor Everolimus revealed that induction of cell death was dependent on sustained engagement of the integrated stress response (ISR)\, resulting in induction of BMF\, a proapoptotic protein of the BCL-2 family. The effects on cell death were phenocopied by the bisteric mTORC1 inhibitor RMC6272 or by genetic silencing of the mTORC1 cofactor Raptor. Genetic silencing of the ISR master regulator ATF4 also phenocopied these effects\, confirming ISR engagement as a critical mediator of cytotoxicity. Conversely\, epigenetic silencing or siRNA-mediated knockdown of BMF conferred resistance to Everolimus\, whereas histone deacetylase inhibition restored BMF expression and drug sensitivity. Combined treatment with Everolimus and Venetoclax elicited potent\, ISR-dependent antitumor activity in vivo. \n\n\n\nThese findings provide insight into the links between mTORC1 and the ISR and furnish proof-of-concept evidence for mTORC1 inhibition and sustained ISR activation as a viable strategy to overcome Venetoclax resistance to in T-ALL. DESCRIPTION:Welcome to the upcoming seminar holdt by Professor Ole Morten Seternes\, Group Leader of the Cell Signalling and Targeted Therapy Research Group\, Department of Pharmacy\, UIT The Arctic University of Norway. \nTittle: The dual specificity MAP kinase phosphatase DUSP2 acts as distal regulatory node in T Cell signalling \nTime: 22nd October 2025. 14:00 – 15:00 \nPlace: Room B307\, 3rd floor\, Sentralblokken\, Haukeland University Hospital \nAbstract: \nThe dual-specificity MAP kinase phosphatases (MKPs) family plays a critical role in the dephosphorylation and inactivation of various MAP kinases in mammalian cells and tissues. MKPs not only provide a mechanism for spatiotemporal feedback control of these essential signalling pathways but also facilitate crosstalk between distinct MAP kinase pathways and other key signalling modules. The ten mammalian MKPs differ in their substrate specificity and subcellular localization. Several MKPs have been implicated in regulating MAPK signalling in T cells. However\, studies utilizing genetic mouse models have yielded conflicting results regarding which MKP members contribute to this regulation. This prompted us to re-examine the role of MKPs in controlling MAPK signalling using a human T cell model. Our findings suggest that DUSP2 regulates early MAPK activation in these cells and may function as a part of a distal regulatory node in T cell signalling.

DESCRIPTION:13:00 –Assoc. Prof. Morten Rye\, Dept. of Clinical and Molecular Medicine\, NTNU: Spatial Transcriptomics Analysis of Prostate Cancer \nThe cancerous prostate gland is a heterogenous mix of different cancer and normal cells and tissue types\, which is a major source of confounding when large scale molecular – omics analysis is performed on the prostate gland. One way to delineate this heterogeneity is Spatial Transcriptomics\, which can generate individual tissue specific gene expression profiles from thousands of spots placed on a grid across a prostate cancer tissue slice (see illustration). A major clinical challenge in prostate cancer is the ability to distinguish aggressive and non-aggressive prostate cancer in the cancer care pathway\, and Spatial Transcriptomics could be a technology to do more precise molecular diagnostics of prostate cancer. With this in mind\, our Prostomics research group generated spatial transcriptomics datasets (Visium 10x Genomics) on 32 tissue samples from 8 prostate cancer patients\, 5 containing aggressive and 3 containing non- aggressive cancers. Samples were taken specifically from cancer\, cancer-adjacent and distant normal tissue regions\, and spots where classified according to their content of cancer\, extracellular matrix/stroma and normal tissue\, and further analysed by Gene Set Enrichment Analysis. Our analysis discovered that normal appearing tissue adjacent to aggressive cancer regions lose prostate specific functions and adapt properties resembling cell-types which are not expected to be present in a healthy prostate gland. \n  \n13:20 –Prof. Berit Johansen\, Dept. of Biology\, NTNU: Treating cancer and leukemia: A new way to trigger cell self-destruction \nThe polyunsaturated fatty acid arachidonic acid (AA) and its metabolites\, eicosanoids\, i.e. prostaglandins\, thromboxanes\, leukotrienes\, etc.\, have attracted considerable interest in relation to inflammatory processes and diseases\, including cancer (leukemia)\, cardiovascular and autoimmune diseases\, and metabolic disorders. AA is generated by the action of phospholipase A2 (PLA2)\, which catalyzes its hydrolytic cleavage from membrane glycerophospholipids. PLA2 enzymes constitute a superfamily of enzymes\, divided into many groups and subgroups. Among them\, cytosolic Group IVA PLA2 (cPLA2α) plays a critical role in the release of AA and is the most well-studied PLA2 in inflammation. Targeting eicosanoid production by inhibiting cPLA2α offers a promising approach for inflammation or cancer therapy. Our findings imply cPLA2α may support cancer by mitigating oxidative stress and inhibiting tumor suppressor expression suggesting that the very specific and sensitive cPLA2α -inhibitors we have developed\, has potential for treating acute leukemias and other cancers by introducing programmed cell death.

Han var første vinner av Reodorprisen\, har ledet Tomras satsning på radikal innovasjon og vært med på å starte flere selskaper. Han har skrevet åtte innovasjonsfaglige bøker og leder for tiden Norges største innovasjonsfaglige dypdykk\, med 185 deltakere. Sjur vil komme inn på noen grunnleggende sider ved gründerskap\, forskjellen på forskning og innovasjon og hvorfor det enkle ofte er det vanskeligste.
